In this paper, we describe the first co-crystal structure of the protein CYRI-B with its binding partner Rac1. We based on the similarity between CYRI-B and CYFIP to propose a more refined mechanism of how active Rac1 activates the Scar/WAVE complex. We also describe a novel dimerisation mechanism between CYRI molecules as a new regulation mechanism.

By using super-resolution microscopy, we describe the role of a novel protein called CYRI-A in helping resolve the formation of macropinosomes – structures cells use to uptake external substances. CYRI-A achieves this by locally suppressing active RAC1 signalling, therefore regulates how actin polymerises at the macropinocytic cups. By using different 2D and 3D invasion assays, we further show that CYRI protein family also involves in regulating surface integrins in cancer cells through macropinocytosis, and therefore controlling how cancer cells migrate and invade.
